Season Four is set four months after Jordan Collier distributed promicin, the substance that gives the mysterious 4400 their powers, to the world. The world's governments, though unable to stop the distribution or destroy promicin, have outlawed its usage by the population and have ordered NTAC, led by new Regional Director Meghan Doyle, to arrest anyone who is Promicin-Positive.

Key Agents Tom Baldwin and Diana Skouris, while doing this to the best of their ability to enforce law and order in the post-chaotic new world, suffer personal problems that push them to their limit, most notably when relatives close to them become P-Positives.

As much as NTAC tries, the war against Collier and the Promicin Movement is steadily floundering and eventually the foundations of paradise are layed...Promise City.
